Discovery of Bacteria

The discovery of bacteria depends on the invention of the microscope. After the invention of the microscope, the following scientists played their role in its discovery.

Leeuwenhoek was the first scientist who observed microbes in 1673. These microbes were bacteria and protozoa. He used a simple microscope to describe them. He gave accurate drawings and descriptions of these small creatures. He called them animalcules. He firstly observed these small creatures in rainwater. Then he confirmed these observations in saliva vinegar, infusion, and other substances.

Pasteur made many discoveries in microbiology and medicine. He developed vaccines for anthrax, fowl cholera, and rabies. He discovered the process of pasteurization and fermentation. He proved that microorganisms could cause disease.

Germ Theory of disease of Robert Koch

Robert Koch formulated the ‘germ theory of disease’. He isolated a typical rod-shaped bacterial structure from the blood of a sheep that died of anthrax. These bacteria have squarish ends and are called bacilli. Then he discovered the bacteria of tuberculosis (TB) and cholera. He formulated four postulates of his germ theory of disease. These postulates help to understand whether the organism present in the lesion has caused or not caused the disease.Robert Koch

  1. A specific organism can always be found in association with a given disease.
  2. The organism can be isolated and grown in pure culture in the laboratory.
  3. When this pure culture is inoculated into a healthy animal, it produces disease.
  4. The organism can be recovered in pure culture from the experimentally infected animal.

Koch and his colleagues invented many techniques of inoculation, media preparation, maintenance of pure culture, and preparation of specimens for the microscope

Occurrence

They are widely distributed. They are found almost everywhere. The number and kind of bacteria depend on the environmental condition of the locality.

  1. These are present in air, land, water, oil deposits food, decaying organic matter, plants, man, and animals.
  2. They are always present in natural flora.
  3. Some of them are present in specific environments like hot springs alkaline/acidic soil, highly saline environments, in highly polluted soil and water.

Growth in Bacteria

In bacteria, there is very rapid growth. It depends upon suitable temperature, nutrition, pH, and the presence of certain ions. There are different stages of growth in bacteria.

(1) Lag Phase (2) Log Phase (3) Stationary Phase (4) Decline Phase or Death Phase

LAG PHASE:Growth in Bacteria

This is the initial and inactive phase of growth. This is a preparatory stage. The bacteria prepare themselves for reproduction and accumulate certain substances like water and proteins which help in growth.

LOG PHASE:

It is the most active phase of growth. The bacteria increase their number very rapidly and their metabolic activities are maximum. Their rate of growth is rapid and the rate of death is minimum.

STATIONARY PHASE:

When there is a shortage of food, the growth of bacteria becomes slow. There is an equal rate of reproduction and death of bacteria. So, the number of bacteria almost remains constant i.e. no increase and no decrease. Due to an unchanged number of bacteria, the stage is called the stationary phase.

DECLINE OR DEATH PHASE:

When there is an acute shortage of available food, the growth of bacteria is very slow and the rate of death is rapid, their number decreases rapidly, so it is a decline phase. Ultimately they disappear completely from that place, and then it is called the death phase.

Locomotion in Bacteria

In bacteria, locomotion takes place by Flagella. There are fine threads-like structures present on the outer surface of the bacterial cell. Some bacteria have a single flagellum and some have many flagella. Mostly Bacilli bacteria contain flagella and Cocci do not contain flagella. Bacteria usually move towards their food material. According to their movement, there are three types of bacteria.

1.      Phototactic Bacteria

These bacteria move due to light, either towards or away from light.

2.      Chemotactic Bacteria

These bacteria move due to any chemical substances, present in their surroundings. They usually move away from chemical substances.

3.      Magnetotactic Bacteria

These bacteria move due to the presence of a magnetic field, away from the magnetic field.

Nutrition of Bacteria

Bacteria need energy for their growth, maintenance, and reproduction. They get this energy from food. There are two methods of reproduction in bacteria:

1. Heterotrophic bacteria

Most of the bacteria are heterotrophic. They cannot synthesize their organic compounds from simple inorganic substances. They may be saprophyte or parasites:

Saprophytic Bacteria: They get their food from dead organic matter. Soil contains humus. This hummus is full of organic compounds. Humus is materially formed as a result of the partial decay of plants and animals. Many soil-inhabiting bacteria have a very extensive enzyme system. They secrete these enzymes in the humus. These enzymes break down the complex substance of the humus into simple compounds. The bacteria can absorb and use these substances for obtaining their energy.

Parasitic Bacteria: These bacteria obtain their food from the body of living organisms. These are fully dependent on their hosts for their nutrition. These bacteria cause diseases in the host and sometimes death occurs.

2. Autotrophic Bactria

Some bacteria are autotrophic. The bacteria which utilize CO2 and get energy from sunlight or from some chemical reactions are called autotrophic bacteria. They can synthesize organic compounds from inorganic substances. There are two types of autotrophic bacteria:

Photosynthetic Bacteria:

They possess chlorophyll. Their chlorophyll is different from the chlorophyll of green plants. The chlorophyll of the green plants is present inside the chloroplast. But the chlorophyll of bacteria is dispersed in the cytoplasm. The autotrophic bacteria use hydrogen sulfide (H2S) as a hydrogen source, instead of water (H20). They release sulfur instead of oxygen.reaction

There are three types of photosynthetic bacteria. These are:

  1. Green sulfur bacteria.
  2. Purple sulfur bacteria.
  3. Purple non-sulfur bacteria.

Chemosynthetic Bacteria:

They oxidize inorganic compounds like ammonia, nitrates, nitrites, sulfur, or ferrous iron. Energy is released during this reaction and bacteria trap this energy and use it for their activities. Nitrifying bacteria are also chemosynthetic bacteria.reaction

3. Symbiotic Bacteria

Some bacteria form an association with the other organism and both get benefits from each other, such bacteria are called symbiotic bacteria. For example, Rhizobium bacteria live in the roots of leguminous plants (e.g. pea plants). These bacteria convert Nitrogen into its compounds, Nitrates, and Nitrites. This process is called Nitrogen Fixation. As a result of this process, bacteria get food material from plants.
